Pierre Spies moves from the bench into Rossouw's spot, while veteran Bob Skinstad is likely to win his first cap since 2003, when he played his 34th and last Test against Argentina.
"I certainly didn't feel like I'd be here, but now I want to do it to the best of my ability," said Skinstad, who had a spell with Newport.
"It was a near-impossible task to come back after so many years out, but there were so many people at the Sharks that helped me make the transition smooth.
"Guys like John Plumtree, Dick Muir and Smittie (John Smit) were superb."
