The 23-year-old Sale Sharks forward replaces Wasps flanker Joe Worsley, who is sidelined because of a hamstring injury.
It is the solitary change made by head coach Sir Clive Woodward following England's 50-9 victory over Italy last weekend in their RBS 6 Nations Championship opener.
Jones went on as a second-half substitute at Stadio Flaminio, scoring the last of England's seven tries.
"We have been very excited about him (Jones) for the last 12 months, and the guys up at Sale - Jim Mallinder and Steve Diamond - have done a great job with him," said Woodward.
"He is kind of unique, in that he has got this incredible pace, and yet he is six foot, six inches tall. He is very tough and we are looking forward to him playing.
"He adds a lot to our lineout. To go in with Danny Grewcock, Ben Kay and Chris Jones as your specialist lineout forwards, and add to that Lawrence Dallaglio and Richard Hill, and you have a formidable lineout building here.
"Jones is a very special talent - you can't compare him with any other player."
Jones' place on the bench is taken by his Sharks team-mate Alex Sanderson.
Scrum-half Andy Gomarsall has recovered from a thigh strain to retain the number nine jersey, while skipper Lawrence Dallaglio and centre Jason Robinson also feature after suffering minor hand injuries in Rome.
