The other changes come in the pack where Ben McCalman comes in for Wycliff Palu at No.8 and Tatafu Polota-Nau makes his 50th international appearance at hooker.
Kurtley Beale stays on the bench with Bernard Foley preferred at fly-half after his match-winning conversion last time out against South Africa.
Ashley-Cooper is only one game away from his 100th Wallabies Test but that appears likely to have to wait until the clash with the All Blacks next month.
"Tatafu has been a true servant to the game of rugby in Australia for a long time and I couldn't be more pleased to see him earn his milestone 50th game," McKenzie said.
"I've known Tatafu for a long time and he is a guy who puts his body on the line each and every week for the benefit of the team.
"He gives it his all every time he takes the field and is also an outstanding teammate off it."
